Well, seeing as I have to do my least favorite chore, a place I applied at called today and I missed it, my mom is as of right now knocked up on who knows what for her surgery (Or possibly undergoing surgery), and today was the last day I'll see my older brother in awhile (8 months, at the very least), not so good.
Though I did get some pepper jack cheese Tornados, a type of food found in certain gas stations around here, so I'm feeling better.

That's good. I had to go to my grandpas funeral the other day. His aorta ruptured while we were on vacation in South Padre Island.

Bummer. I hate funerals. It's amazing that I've not been to any weddings but I've already been to two or three funerals o3o
I forgot to mention I'm also stuck at home with two crazy cats, a bitchy dog, a stinky dog, my younger brother, and my dad.

You don't need to make a second post to tell us something we can find out by going to the main part of the forum.

Congratulations on getting the job!
A few recommendations from someone who has two right now...
1. For the first month or so, remember that you're new and it's OKAY to ask questions. Better you ask your supervisor how to do something or what to do than mess up, and have them have to fix stuff later.
2. Be an overachiever. If you've finished all the stuff they've told you to do - go find your supervisor and ask for more. It gives a really good impression, and then you start figuring out the little things you can do on your own that really help. Once you know your supervisors and your co-workers a little better you can figure out how much you should do - etc. but at the beginning it's good to be extra helpful.
3. Your experienced co-workers are your best friends - be nice to them, and they'll show you the tricks of the trade.
4. Leaving early and showing up late are no-no's the first month or so. Once you know your supervisor/coworkers a little better you can feel this out for yourself, but at first don't risk it.
Just things I've found out over the years. Especially number one (I accidentally killed a cell line from Saudi Arabia that way and we had to fly in new sample cells. SUPER EMBARRASSING!). Of course I'm on at work right now - but it's because most processes in cell biology are: do stuff for an hour. Wait four. Do stuff for an hour. Wait four... etc. I'm in the wait four section right now.

Yeah - four hours - though since I put them in at 2 hours ago - it's only two now.
Basically - when you freeze cells you first put them into a serum that will shock them into protecting themselves (a poison basically). This makes it so that when you freeze them at -80C or so they are already protecting themselves. But when you thaw them - you have to let the cells get settled, but at the same time you have to deal with the poison. So you dilute it at first with new growth media - then four hours later when the cells are all nice and settled you wash them down with new media to make sure you've got all of the poison off. Simple enough - but TONS of waiting involved...
Basically - when you freeze cells you first put them into a serum that will shock them into protecting themselves (a poison basically). This makes it so that when you freeze them at -80C or so they are already protecting themselves. But when you thaw them - you have to let the cells get settled, but at the same time you have to deal with the poison. So you dilute it at first with new growth media - then four hours later when the cells are all nice and settled you wash them down with new media to make sure you've got all of the poison off.
